Sanctify level is completely unrelated to Star level. Since the patch which changed it, the medal cost increases as you gain Star levels.

But does the frequency of the stars taking have anything to do with the level of the hero? Also are they pretty much predetermined? Or does the chance to hit or not change daily with server reset?

The only things that matter in determining the number of medals needed is star level and color of the hero.
